I believe you cannot "read" it while it is running. So the way to do it ( the way i know of ) is:
- Load up linux on a live CD
- Copy the whole Folder which contains .SAM and some registery things to a memory stick
- Load up windows again
- Download a .SAM cracking tool ( the one i use is SAMinside )
- Open SAMinside and import the copyed .SAM file and registery file
- Let it start Brute Forcing.
There are others ways to do it i belive such as using a .SAM cracker in Linux so you dont have to copy the SAM file.

First of all, their is a repair version that can be used whilst the actual System32 one is running C:\WINDOWS\repair\SAM
And second as they said use a program that reads the SAM (Security Accounts Manager) file hash and cracks it, simple their are many tools out their to do just that :)
